English Analisis In-silico Senyawa Bioaktif dari Pakis Tanduk Rusa (Platycerium bifurcatum C. Chr.) sebagai Kandidat Obat Anti-Inflamasi untuk Menghambat Enzim Siklooksigenase-2

Abstrak

Inflamasi dapat ditekan melalui mekanisme antiinflamasi dengan cara menghambat kinerja enzim siklooksigenase-2 yang berakibat pada menurunnya sintesis prostaglandin sehingga tanda-tanda klinis inflamasi dapat dikurangi. Mekanisme antiinflamasi dapat dilakukan oleh kelompok obat NSAID seperti asetetasin, namun penggunaan secara terus-menerus dapat menimbulkan efek samping sehingga membuka jalan bagi pemanfaatan bahan alam sebagai pengobatan alternatif seperti daun P. bifurcatum C. Chr. Penelitian ini bertujuan untuk menganalisis potensi senyawa bioaktif pada daun P. bifurcatum C. Chr sebagai antiinflamasi berdasarkan nilai afinitas ikatannya dengan siklooksigenase-2, potensial biooral, ADMET, dan uji toksisitas melalui pendekatan in silico dengan teknik molecular docking. Hasil penelitian menunjukkan bahwa pada daun P. bifurcatum C. Chr terdapat senyawa asam galat dan quercetin. Nilai afinitas pengikatan kontrol asetamin (kontrol) adalah -7,9 kkal/mol, asam galat -6,3 kkal/mol, dan quercetin -8,8 kkal/mol. Acemetacin (kontrol), asam galat, dan quercetin mengikat enam residu asam amino identik dari siklooksigenase-2, yaitu THR A:212, TYR A:385, HIS A:388, TRP A:387, ALA A:199, LEU A:390. Quercetin dan asam galat dapat diserap, didistribusikan, dimetabolisme, dan diekskresikan dan tidak menunjukkan risiko kesehatan apa pun. Keduanya dapat dikonsumsi secara oral dengan aman dan tidak menyebabkan kanker dalam jangka panjang, sehingga dapat menjadi kandidat obat antiinflamasi.
