Abstrak

Kanker merupakan penyakit peringkat ke enam sebagai penyebab kematian terbesar di dunia pada tahun 2016. Prevalensi kanker di Indonesia dari tahun 2013 hingga 2018 meningkat sebanyak 0,6%. Metode pengobatan yang ada saat ini menimbulkan efek samping yang tidak diinginkan, sehingga banyak masyarakat kembali pada pengobatan herbal. Senyawa D-alpha-Tocopherol yang terkandung dalam tanaman zodia (Evodia suaveolens) yang merupakan tanaman endemik Papua memiliki sifat antioksidan yang baik untuk antikanker. Penelitian ini bertujuan untuk mengetahui efektivitas dan bioaktivitas senyawa D-alpha-Tocopherol sebagai antikanker pada kanker paru-paru melalui protein EGFR (Epidermal Growth Factor Reseptor) dan kanker kolorektal melalui protein ALK (Anaplastic Lymphoma Kinase) secara in silico. Metode penelitian yang digunakan yaitu biokomputasi secara in silico, meliputi koleksi protein dan senyawa, sterilisasi dan isolasi protein, minimasi senyawa, docking, dan visualisasi hasil docking antara protein dengan senyawa. Hasil penelitian menunjukkan bahwa senyawa D-alpha-Tocopherol berikatan dengan protein EGFR dengan binding affinity -4,6 KJ/mol; RMSD 1,760 Ã…. Ikatan senyawa D-alpha-Tocopherol dan protein  ALK dengan nilai binding affiniy -7,6 KJ/mol; RMSD 2,167. Tingginya nilai binding affinity dan RMSD menunjukkan bahwa penggunaan senyawa D-alpha-Tocopherol sebagai obat antikanker kurang efektif. Sifat antioksidan D-alpha-Tocopherol memberikan keuntungan bagi sel kanker untuk tetap hidup.
